No. A serial port on a Core does not require a network port to be active to actually operate with AMX/Crestron.
Serial ports in Q-SYS are not dedicated to a particular function so to provide specific functionality to operate with AMX/Crestron, the serial port block on the Core needs to be attached to a script-based component that contains the desired functionality.

It is the requirement of the script-based component, what it does and how it operates that determine if a network connection is required, not the serial port itself.
